PNC-27 contains the MDM-2 binding domain of p53 along with a transmembrane penetrating domain. In cancer cells, which overexpress HDM-2 on their membranes, PNC-27 binds HDM-2 and forms pores that lyse the cancer cell. Normal cells do not express membrane HDM-2 and are unaffected. This selective mechanism distinguishes PNC-27 from conventional cytotoxic approaches and represents a fundamentally novel anticancer peptide strategy.
